Edificado entre 1527 y 1540, el Museo Regional de Tlaxcala está ubicado en un ex convento franciscano del siglo XVI. El lugar ha protagonizado diversos acontecimientos y ha tenido diferentes usos, como hospital e incluso como cárcel. Fue inaugurado como museo en 1985.
